GR-04 A novel regulatory network of left–right asymmetry establishment in Drosophila melanogaster: Interaction between the unconventional myosins ID and IC and the adherens junction component DE-cadherin

چکیده
somitogenesis clock was evaluated by assessing the rate and number of somites formed and gene expression alterations upon notochord ablation. Chick embryo explants were cultured for different time periods with or without notochord. We found that notochord removal delays somite formation from the undetermined PSM and alters both clock and determination front gene expression. We show that Shh is the notochord-derived signal responsible for this effect since the notochord ablation phenotype is recapitulated by the use of Shh pathway inhibitors and rescued by the juxtaposition of Shh-producing cells. Moreover, we show that Shh is regulating the period of clock oscillations. Altogether, our data strongly suggest that embryo midlinederived Shh is overseeing somitogenesis clock periodicity and timely somite formation.

DE-Cadherin regulates unconventional Myosin ID and Myosin IC in Drosophila left-right asymmetry establishment.
In bilateria, positioning and looping of visceral organs requires proper left-right (L/R) asymmetry establishment. Recent work in Drosophila has identified a novel situs inversus gene encoding the unconventional type ID myosin (MyoID). In myoID mutant flies, the L/R axis is inverted, causing reversed looping of organs, such as the gut, spermiduct and genitalia. We have previously shown that Myo...
متن کاملLeft-right asymmetry: class I myosins show the direction.
Myosins are actin-based molecular motors that are found in almost all eukaryotes. Phylogenetic analysis allows the discrimination of 37 different types of myosins, most with unknown functions. Recent work in Drosophila has revealed a crucial role for type ID unconventional myosin in left-right asymmetry. Mutations in Myosin ID completely reverse the left-right axis (situs inversus), a phenotype...
متن کاملRho1 regulates adherens junction remodeling by promoting recycling endosome formation through activation of myosin II
Once adherens junctions (AJs) are formed between polarized epithelial cells they must be maintained because AJs are constantly remodeled in dynamic epithelia. AJ maintenance involves endocytosis and subsequent recycling of E-cadherin to a precise location along the basolateral membrane. In the Drosophila pupal eye epithelium, Rho1 GTPase regulates AJ remodeling through Drosophila E-cadherin (DE...
متن کاملE-Cadherin Is Required for Centrosome and Spindle Orientation in Drosophila Male Germline Stem Cells
Many adult stem cells reside in a special microenvironment known as the niche, where they receive essential signals that specify stem cell identity. Cell-cell adhesion mediated by cadherin and integrin plays a crucial role in maintaining stem cells within the niche. In Drosophila melanogaster, male germline stem cells (GSCs) are attached to niche component cells (i.e., the hub) via adherens jun...
متن کاملAconitase and Developmental EndPointsasEarly IndicatorsofCellularToxicity Induced by Xenobiotics in Drosophila Melanogaster
Background: In this study, the toxicity of the different xenobiotics was tested on the fruit fly Drosophila melanogaster model system. Methods: Fly larvae were raised on food supplemented with xenobioticsat different concentrations (sodium nitroprusside (0.1-1.5 mM), S-nitrosoglutathione (0.5-4 mM), and potassium ferrocyanide (1 mM)). Emergence of flies, food intake by larvae, and pupation h...
